    We are proud to announce the publication of the world's first integrative neurodevelopmental model describing the mechanisms and dynamics of ASD pathogenesis from pregnancy to child early life: "Autism Spectrum Disorders pathogenesis: Toward a comprehensive model based on neuroanatomic and neurodevelopment considerations". the latest output of the "GEMMA (Autism & microbiota) H2020 program" launched in 2019.
    This is the 2nd in a series of 4 publications with Harvard Medical School. The first presented a novel mechanistic hypothesis on the interaction between the microbiome and the host in ASD affected individuals: "Autonomic Nervous System Neuroanatomical Alterations Could Provoke and Maintain Gastrointestinal Dysbiosis in Autism Spectrum Disorder (ASD): A Novel Microbiome–Host Interaction Mechanistic Hypothesis". The third will further explain the relative weight of epigenetics and RNA editing mechanisms in the development of ASD, while the fourth will conclude the overall neurodevelopmental model.

    Summary
    Taking advantage of the accessibility of the skin in vivo, non-invasive methods were developed about 40 years ago, which have now developed as accurate sources to measure skin properties and structures at the microscopic and macroscopic levels. However, even at the dawn of the twenty-first century, the mechanisms involved in these properties are still only partly understood. As in many domains, including biomedical engineering, numerical modeling has appeared as a complementary key actor for improving our knowledge on skin physiology.
    This book gathers, for the first time, chapters describing scientific computing and numerical modeling for a deeper understanding of mechanisms involved in skin physiology. The book is structured around some skin properties and functions, including optical and biomechanical properties, skin barrier function, and homeostasis, covered through several chapters describing either biological or physical models at different scales.

    Abstract: Cognitive disorders are highly heterogeneous in terms of symptoms, clinical aetiologies, disease progression and thera- peutic responses. Furthermore, their potential biological causes remain largely unknown. Progress at these different levels is cur- rently mired in a vicious circle. The identification of coherent bio- markers, essential for clinical and therapeutic progress, requires an understanding of either the relevant pathogenic processes or, at the very least, of the parameters that need to be monitored. But, syndrome-dominated conceptual thinking has become a barrier to understanding the biological processes linked to diseases characterized by clinical and therapeutic heterogeneity. As a result, current biomarkers of cognitive disorders are much too numerous, too heterogeneous and too variable to serve useful purposes. This leads to an untenable situation that precludes coherent therapeutic developments since it effectively prevents defining what could constitute valid biological, clinical and therapeutic approaches. How to escape from this situation? The problem could be partly resolved by adopting the much wider views allowed by “system-wide” approaches: indeed, by constructing predictive theoretical models of what could constitute pathological cognitive processes.
    This, naturally, shall require the integration of massive amounts of highly heterogeneous and often conflicting information. The following article aims to provide a necessarily brief overview of the concepts, the breadth of data and the variety of network dynamics that will have to be considered while proposing a functional model- building approach, experimentally validated in vitro and in vivo,
    that could be fruitfully utilised.

    The third paper unravels the impact of epitranscriptomics in ASD pathogenesis by explaning how abrupt
    changes in environmental conditions in utero, such as those conferred by maternal inflammation/immune
    activation, influence RNA editing mechanisms, with the combination of these processes altering fetal

    Aepodia have decided to combine their respective competences. Conscious of the urgent needs to significantly improve and accelerate knowledge and treatments in the realm of specific pains, where the challenges areconsiderable and therapeutic responses non satisfactory, the partners have launched an innovative research program which combines the scientific, combined products development and clinical know-how of the research mathematical (CADI™) modeling and scientific talents of BMSystems’ team of biologists. More specifically, Bio-Modeling Systems and Aepodia have started to construct two new CADI™ models that describe and explain the Fibromyalgia and chronic facial pain mechanisms. The purpose of this collaboration is to open new avenues that will be decisive for the understanding, the diagnosis and the treatment of severe pains. The A1 phase of this program is entirely self-financed by the partners, revealing their confidence in their capacity to jointly identify and characterize mechanisms that will lead quicker to specific combined therapies to the patients.

    Provide possible “contact allergy free” formulation strategy combining interesting allergic products with their corresponding inhibitors to develop innovative formulas.
    Develop a range of products/services to fit clients’ needs.

    Bio-Modeling Systems (BMSystems) and the Foundation for Scientific Cooperation FondaMental have decided to merge their respective competences. Conscious of the urgent needs to significantly improve knowledge and treatments in the realm of psychiatry, where the challenges are considerable and therapeutic responses non satisfactory, the partners have launched an innovative research program which combines the scientific and clinical know-how of the research teams attached to the foundation FondaMental and the heuristic, non-mathematical (CADI™*) modeling talents of BMSystems’ team of biologists.

    More specifically, Bio-Modeling Systems and the Foundation FondaMental aim to construct a new CADI™ model that describes and explains the immuno inflammatory mechanisms that can underlie the pathogenesis and clinical progression of bipolar disorders and schizophrenia. The purpose of this collaboration is to open new avenues that will be decisive for the understanding, the diagnosis and the treatment of severe psychiatric disorders. The A1 phase of this program is entirely self-financed by the partners, revealing their confidence in their capacity to jointly identify and characterize mechanisms that will allow adapting treatments to the physiopathological profiles of patients. The scientific program is placed under the shared leadership of Pr. Marion Leboyer, director of the Foundation FondaMental, and Dr. François Iris, scientific director of Bio-Modeling Systems.

    Context: Numerous attempts to utilize low-toxicity metabolic co-factors have been made over the years but all have proven ineffective.

    Objective: Build a CADI™ model that describes the mechanisms which:
    1-could give rise to mitochondrial dysfunctions that would
    2-most severely affect dopaminergic neurons.

    Work done or to be done:
    The IDUNN program led to a combinatorial therapeutic approach* utilizing two molecules that had long been on the market, neither of which has any known toxicity or undesirable effects.
    This potential treatment was exposed, under strict confidentiality, to the criticism of internationally respected clinical specialists. It received their full approval.
    Implementation of the clinical proof of concept upon Parkinson’s disease patients is currently under negotiation.

    DISEASE: Describe the physiopathology of the disease with clinicians and patients feedback.
    MECHANISMS: Discovery of the causal and symptomatic Mechanisms of the disease.
    BIOMARKERS: Identify relevant biomarkers or specific combination of biomarkers that could measure de mechanisms deregulations at different stages of the disease.
    TARGETS: based on the mechanisms, Identify what could be the best targets to specifically address the causative deregulations.
    SOLUTIONS: We “play” with the mechanisms. Propose a diagnostic and therapeutic strategy to address the relevant mechanisms deregulations with existing drugs or novel drugs with the necessary delivery systems and therapies follow-up.
    FEED BACK LOOP: Integrate the results from e R&D or e Health experimentations into the validation process to improve the global patient and disease follow-up.
